227 - DNA replication
P1 - Scientists recognize semiconservative replication as the mechanism by which DNA replicates prior to cell division.
Process: bases of the DH are broken => molecule unzips into two strands => each strand joins with a new one=> each copy is an equal mix of old+new.
Prior to the M experiement of 1958, other mehods:
- conservative replication
- dispersive replication
P2 - M experiment:
Grew E.c bacterial cultures, some with heavy N. isotopes, some with lighter => distinguis between old nand new
1/ the bacteria grown in Heavey transferred to medium containing the lighter DNA
2/ after one generation, intermediated with half heavy half light
3/ after two, half intermediate, half light
Semiconservative replication also occurs in eukaryotes, such as human chromosome.
P3 - When W and C deciphered the DNA structure in 1953 they intuited the semiconservative model was correct.
Still a clever guess until M and S devised away to differentiate between the original strands of DNA and future generations.
Autoradiographic images confirmed both W&C's intuition and M&S's experiement.
