If they know you equally well, the higher one could be better. However usually direct supervisor knows you more.

Here, the key is to go with the person who knows you best.  The person’s title is less important than selecting the person who can speak with the most personal knowledge.  If you can find someone who knows your work well, and someone who will be open to helping you, then THAT is your guy.  If the two people know you equally as well, and if the two people can speak equally knowledgeably about your work, THEN you can pick the person with the higher title.  But that is less important than picking the person who knows you best, and the person who will fight hardest for your application!  J
