I don't really have enough info from you to tell you whether you have a chance at Darden or not. Two years of work experience when you apply (which means three years when you matriculate) can be enough to apply certainly, but it's more than just the fact or working two years or not that is important. What is important also is WHAT you have done in those two years. Also you may want to retake the GMAT (what score did you get BTW?) and the TOEFL to get the best score possible for you. Keep in mind that you will only do this ONCE, so it's best not to rush things, and to write your applications with the highest possible score in hand.
Sure. Make sure you get a good GPA score to show them you're smart. With a poor GPA though, the concern is 'smart but lazy'. With good social activities, that can go someway to explain the GPA... An optional essay would be a good idea.
