Marconi's conception of the radio was as a substitute for the telephone, a tool for private conversation; instead, it is...

I already know that it is unclear what "it" refers to, therefore this answer is incorrect;
but can "it" refers to "radio" in the prepositional structure "of..." ?

一般情况下不能指代介词后面的宾语。
如果主要成分里面有符合的优先指代它啦~

No. "of the radio" is a prepostional phrase. If you eliminate all prepositional phrase and modifiers, you will see the CORE SENTENCE which is Marconi's conception was as a substitute; instead, it is...

Therefore, it can not refer radio.
